Isolating unidentified bacteria from the rhizosphere to identify them and subsequently show recruitment of these bacteria.
To isolate the bacteria from the rhizosphere I utilized several bacterial agar media. I obtained pure cultures with the use of dilution series and sequenced the 16s rRNA of the bacteria with the use of PCR amplifications. Then I identified the bacteria with BLAST tools. For the bacteria of interest I formed natural rifampicin resistant strains and used them for a recruitment experiment. I statistically compared the results of this experiment with ANOVA tests.
Universiteit Utrecht February 2013 - December 2013Title: Heterogeneity of Aspergillus niger grown in bioreactors
The translation of results obtained in laboratorial liquid shaken cultures to industrial bioreactors and to research the heterogeneity of Aspergillus niger in bioreactors.
Firstly I had to optimise the growth conditions of the small scale bioreactors (2L). After optimization I grew various Aspergillus niger strains in the bioreactors, including fluorescent strains. I measured and analyzed the size, optical density and fluorescence of the fungi with the COPAS, the programs R and Scilab, the light microscope and the confocal microscope.
Additionally I worked on creating a knock-out strain. I used the program CloneManager to design the construct and then created it with PCR, gel-electrophoresis, restriction reactions and ligation reactions. I reproduced the plasmid in competent E. coli cells and then transformed self-made Aspergillus niger protoplasts with the plasmid.
