The Position

Purpose: Optimizing Vector Design, Transgene Delivery, and Primary T-Cell Expansion for Cell

Engineering and Cell Therapy Purposes


We are looking for a motivated and exceptional postdoctoral research scientist for at scale optimization of steps involved in personalized T-cell engineering process. After isolation and enriching of patient’s T-cells, the endogenous TCR of these cells need to be deleted and replaced with tumor specific TCR. Properly targeted T-cells should then be cultured and expanded in order to be reintroduced back to the patient. However, protocols and approaches for at scale implementation of many of these critical steps are suboptimal and require highly innovative and extensive improvements to make the therapeutic approach efficient and cost effective. The successful postdoc candidate would be involved in at scale optimization of vector design, transgene delivery, and high TCR transgene knock in (KI) and endogenous TCR knock out (KO) in primary T cells. The work involves testing a variety of non-viral DNA vector designs, transfection reagents, and approaches that enhance homologous recombination rates to develop a robust protocol for T-cell engineering. Functionality of the derived engineered T-cells would be tested using multiplex characterization devices. Other aspects of this project involve screening small molecule libraries to identify compounds that can enhance homologous recombination rates or modulate T-cell growth and expansion in vitro, and understanding the cellular pathways and proteins that are utilized by adeno-associated virus (AAV) making this mode of gene delivery so efficient. The expected outcomes are discoveries that significantly improve autologous T-cell engineering and publication(s) in scientifically sound and peer-reviewed journals.


The ideal candidate will have expertise in autologous and/or CAR-T cell engineering, molecular/cellular biology, and biochemistry. Familiarity with standard molecular biology techniques, DNA manipulation and cloning, western blotting, PCR/qPCR, gene transfection, recombinant protein expression/analysis, and cell culture techniques are requirements. The candidate must be a good communicator with strong presentation skills and the ability to investigate and convey complex and difficult ideas and data both verbally and in writing. Expectations for this position include: very strong work ethic, being highly collaborative, initiation and completion of investigative projects within the expected time frames, and with emphasis on accuracy and high-quality work.

A PhD in a related field with 0-2 years of postdoc experience is required. Qualified candidate is expected to have first author publications in related fields (biochemistry, immunology, cellular/molecular biology, or cancer biology).
